异夏佛塔苷对人非小细胞肺癌A549细胞抗肿瘤的活性及其作用机制
The anti-tumor activity and mechanism of isoschaftoside on human A549 cell lines
【摘要】 目的探讨不同浓度异夏佛塔苷(Isoschaftoside)对人非小细胞肺癌A549细胞抗肿瘤活性及其作用机制的影响。方法采用MTT法检测不同浓度的异夏佛塔苷对A549细胞增殖的作用;采用Annexin V FITC/PI双染法流式细胞术检测异夏佛塔苷对A549细胞凋亡的影响;蛋白印迹法(Western blotting)检测异夏佛塔苷对A549细胞凋亡相关基因Bax、Bcl-2和Caspase-3表达的影响。结果 MTT结果显示:与对照组相比,异夏佛塔苷对A549细胞有明显的抑制增殖作用,且呈时间剂量依赖性;流式双染检测显示:异夏佛塔苷作用于A549细胞48h后可增加细胞的凋亡率,且作用具有剂量依赖性;Western blotting显示:异夏佛塔苷可上调Bax、Caspase-3基因的表达,下调Bcl-2基因的表达。结论异夏佛塔苷在体外具有明显的抗肿瘤活性,其机制可能与上调Bax、Caspase-3基因的表达,下调Bcl-2基因的表达有关。
【Abstract】 Objective To investigate the antitumor activity and its mechanism of isoschaftoside on human A549 cell lines(human non-small-cell-lung cancer cells). Methods The MTT assay was used to determine the effect of isoschaftoside at different doses on A549 cells proliferation. The apoptosis of A549 cell line was evaluated by flow cytometry.
