BCA法优化补骨脂蛋白质的提取工艺

【摘要】 采用碱提酸沉法提取补骨脂蛋白质,以二喹啉甲酸(BCA)法测定的蛋白质溶出率为指标,通过单因素实验和正交实验优化补骨脂蛋白质的碱提工艺条件,考察浸提p H、料液比、温度、时间等因素对补骨脂蛋白质提取的影响.结果表明,四种提取因素的影响从大到小的顺序为:浸提p H>料液比>温度>时间.最佳工艺条件为浸提p H 10.0、料液比(g/m L)1∶20、温度45℃、浸提时间150 min,此条件下补骨脂蛋白质溶出率为23.69%.以BCA法测定酸沉蛋白质浓度,确定补骨脂蛋白质的最大沉淀量p H为4.5,蛋白质的最大沉淀率为55.16%.对比发现理论提取率与实际提取率基本相同,证明BCA法测定补骨脂蛋白质的方法准确、可靠,为补骨质蛋白质的进一步研究利用奠定了基础.

【Abstract】 The extraction rate which measured by BCA method was applied to evaluate the protein extraction from Psoralea corylifolia by alkali extraction and acid precipitation method. Both of single factor experiment and orthogonal test were used to optimize the four parameters of alkali extraction technology. Results show that the most important factor is the extraction PH,then is the ratio of solid to the liquid,the extraction temperature,and the extraction time. The most suitable extraction conditions for Psoralea corylifolia protein were as follows: suitable extraction p H is 10.0; ratio of material to the liquid is1 ∶20; extraction temperature is 45℃ and the extraction time is 150 min. Under the determined extraction conditions,protein dissolving rate is 23.69%. As the same,the acid precipitation rate under different p H obtained by BCA method was compared,and the best precipitation rate p H is 4.5,and the largest precipitation rate is 55.16%. In the end,by comparing theoretical protein extraction rate with the practical protein extraction rate,the research found that BCA method was creditable.
